A histological study on the development of the digestive system of Pseudosciaena crocea larvae and juveniles ArchiMer
Mai, Kangsen; Yu, Hairui; Ma, Hongming; Duan, Qingyuan; Gisbert, Enric; Zambonino, Jose-luis; Cahu, Chantal.
The ontogenetic development of the gut and accessory organs in large yellow croaker Pseudosciaena crocea was investigated using light microscopy from hatching up to the juvenile stage (40 days post hatch, dph). At 3 dph (mean +/- S.D., 4.1 +/- 0.1 mm total length, L-T), coinciding with the buccopharynx opening, larvae started to feed exogenously, and the gut consisted of a well-developed buccopharynx, a partially-differentiated oesophagus and an intestine divided in three regions (anterior intestine, intermediate intestine and rectum). Yolk reserves were not completely depleted at the onset of exogenous feeding, and a period of mixed nutrition was observed up to 6 dph (4.3 +/- 0.1 mm L-T) when yolk was definitively exhausted. Important morphological...

Activities of selected digestive enzymes during larval development of large yellow croaker (Pseudosciaena crocea) ArchiMer
Ma, Hongming; Cahu, Chantal; Zambonino, Jose-luis; Yu, Hairui; Duan, Qingyuan; Le Gall, Marie-madeleine; Mai, Kangsen.
Large yellow croaker is an important marine fish species for aquaculture in China. Larval rearing of this species is still based on live prey feeding. The aim of this study was to describe the onset and development of the main digestive enzymes during larval development of this species, in order to obtain essential data for the formulation of a compound diet adapted to larvae. Enzymatic assays were conducted from day I post hatching to day 40 in larvae fed the following live prey sequence: rotifers, Artemia nauplii, copepods, and then frozen copepods. Assays were done on whole larvae until day 19, then on dissected larvae and on purified brush border membranes of intestine. Pancreatic enzymes, amylase, and trypsin, showed high activities from early stages,...

Allometric growth patterns and development in larvae and juveniles of thick-lipped grey mullet Chelon labrosus reared in mesocosm conditions ArchiMer
Ben Khemis, Ines; Gisbert, Enric; Alcaraz, Carles; Zouiten, Dora; Besbes, Raouf; Zouiten, Amina; Masmoudi, Ahmed Slaheddine; Cahu, Chantal.
Allometric growth and ontogeny were studied in thick-lipped grey mullet Chelon labrosus reared in mesocosms from 1 to 71 day post hatching (dph). Multivariate allometric analysis of morphometric growth distinguished three distinct developmental stanzas separated by two morphometric metamorphosis lengths (L-m1=4.46 +/- 0.06mm; L-m2= 28.56 +/- 1.04mm). Body mass growth also showed three distinct episodes separated by two inflections, correlated with morpho-functional changes. First episode concerned pre-flexion larvae and ended around 4.5mm-L-T (14-dph), coinciding with estimated L-m1. It was distinguished by reduced growth, but intense morphogenesis and differentiation processes. Organogenesis and allometric changes indicated that development priorities...

Ocean warming, eutrophication and consequent decrease in oxygen lead to smaller average fish size. Although such responses are well-known in an evolutionary context, involving multiple generations, it appears to be incompatible with current rapid environmental change. Rather, phenotypic plasticity could provide a means for marine fish to cope with rapid environmental changes. However, little is known about the mechanisms underlying plastic responses to environmental conditions that favour small phenotypes. Our aim was to investigate how and why European sea bass that had experienced a short episode of moderate hypoxia during their larval stage subsequently exhibited a growth depression at the juvenile stage compared to the control group. We examined...

The aim of this study was to determine biofloc contributions to the antioxidant status and lipid nutrition of broodstock of Litopenaeus stylirostris in relationship with their reproductive performance and the health of larvae produced. Shrimp broodstock reared with Biofloc technology (BFT) compared to Clear water (CW) exhibited a higher health status with (i) a better final survival rate during the reproduction period (52.6% in CW against 79.8% in BFT); (ii) higher glutathione level (GSH) and total antioxidant status (TAS), reduced oxidized/reduced glutathione ratio and a higher spawning rate and frequency as well as higher gonado-somatic index and number of spawned eggs. Finally, larvae from broodstock from BFT exhibited higher survival rates at the Zoe 2...

Biofloc technology applied to rear shrimp Litopenaeus Stylirostris broodstock: An integrated and development research project in New Caledonia and French Polynesia ArchiMer
Chim, Liet; Cardona, Emilie; Lorgeoux, Benedicte; Gueguen, Yannick; Saulnier, Denis; Goguenheim, Jean; Wantiez, Laurent; Cahu, Chantal.
Shrimp farming in New-Caledonia faces up to difficulties. A fluctuating quality of broodstock prevents sustainable production of larvae in quantity and quality. The traditional extensive rearing method of broodstock in outdoor earthen ponds used in New Caledonia raises several issues: poor water quality control and biosecurity. Therefore it is fundamental to modify the broodstock culture strategy in order to achieve an easier water quality management and maximal biosecurity. Biofloc technology (BFT) offers easier water quality management, higher natural productivity, higher level of biosecurity, and could be a good alternative. Since 2011 a R&D program started under the supervision of IFREMER in New Caledonia and French Polynesia in order to study and...

Characteristics of fads2 gene expression and putative promoter in European sea bass (Dicentrarchus labrax): Comparison with salmonid species and analysis of CpG methylation ArchiMer
Geay, Florian; Zambonino, Jose-luis; Reinhardt, Richard; Kuhl, Heiner; Santigosa, Ester; Cahu, Chantal; Mazurais, David.
Marine fish species exhibit low capacity to biosynthesise highly unsaturated fatty acid (HUFA) in comparison to strict freshwater and anadromous species. It is admitted that the Delta(6) desaturase (FADS2) is a key enzyme in the HUFA biosynthetic pathway. We investigated by quantitative PCR the relative amounts of FADS2 mRNA in European sea bass (Dicentrarchus labrax) in comparison with a salmonid species, the rainbow trout (Oncorhynchus mykiss L.). The analysis of the expression data was performed regarding the difference of the characteristics of a critical fragment of the fads2 gene promoter between sea bass and Atlantic salmon. The lower level of fads2 gene expression observed in sea bass suggested that fads2 gene putative promoter, which exhibited an...

Cloning of endothelin-1 (ET-1) from European sea bass (Dicentrarchus labrax) and its gene expression analysis in larvae with retinoic acid-induced malformations ArchiMer
Geay, Florian; Darias, Maria J.; Santigosa, E.; Desbruyeres, Elisabeth; Quazuguel, Patrick; Zambonino, Jose-luis; Cahu, Chantal; Mazurais, David.
It is known in vertebrates that endothelin-1 (ET-1) plays a key role ill morphogenesis whose expression level greatly influences the development of craniofacial malformations. In the Present study, the complete cDNA fragment encoding a precursor of endothelin-1, the preproendothelin-1 (PPET1), was cloned by RACE-PCR from European sea bass. The cDNA encoded a 199 amino acid polypeptide that was composed of the "mature" and "big" ET-1. Relative ET-1 expression levels were investigated in European sea bass larvae fed microparticulate diets containing the standard amount (0.08 g retinol/kg diet: group N) or all excess of retinoic acid which induces skeletal malformations (0.5 g retinol/kg diet: group RA). Real-time reverse transcriptor polymerase chain...

Fish are the main source of the n-3 highly unsaturated fatty acids, which are crucial for human health. Their synthesis from C18 precursors is mediated by desaturases and elongases, but the activity of these enzymes has not been conclusively established in marine fish species. This study reports the cloning, tissue expression, and functional characterization of a sea bass (Dicentrarchus labrax L.) Δ6-desaturase and one of its splicing variants. Two cDNAs with open reading frames of 1,346 and 1,354 bp were cloned and named D6D and D6D-V, respectively. Both deduced protein sequences (445 and 387 amino acids, respectively) contained two transmembrane regions and the N-terminal cytochrome b5 domain with the HPGG motif characteristic of microsomal desaturases....

Comparison of growth, digestive system maturation and skeletal development in sea bass larvae reared in an intensive or a mesocosm system ArchiMer
Zouiten, Dora; Ben Khemis, Ines; Masmoudi, Ahmed Slaheddin; Huelvan, Christine; Cahu, Chantal.
The quality of development in intensive or mesocosm hatchery-reared Dicentrarchus labrax larvae was investigated using physiological indicators assessing ontogeny. Larvae were reared in intensive (120 L tanks) and in mesocosm systems (20 m(3) enclosures) with the same feeding sequence, excluding the wild zooplankton naturally available in mesocosms. Faster growth was recorded since early development [16 day after hatching (DAH)] in the mesocosm. Maturation of the digestive system also occurred earlier as indicated by the higher amylase secretion ratios, the intestinal maturation index (alkaline phosphatase/leucine-alanine peptidase and aminopeptidase-N/leucine-alanine peptidase ratios) and the more developed intestinal epithelium at 23 DAH. Nevertheless,...

Dietary lipid level affects fatty acid composition and hydrolase activities of intestinal brush border membrane in seabass ArchiMer
Cahu, Chantal; Zambonino, Jose-luis; Corraze, Genevieve; Coves, Denis.
Triplicate groups of juvenile seabass (initial weight of 241 g) were fed during 13 weeks three isonitrogenous experimental diets containing different lipid levels, 12% (LL group), 21% (ML group) and 30% (HL group). At the end of the experiment, fish weight gain was similar among the 3 dietary groups. Intestinal brush border membranes were purified for each dietary group; one part of the brush border fraction was dedicated to enzyme assays, the remaining fraction being used for lipid extraction followed by fatty acid analysis. The fatty acid composition of the brush border membrane differed among the 3 groups, although the 3 experimental diets had the same fatty acid composition. The increase in dietary lipid level resulted in a lowering in (n-3)...

Dietary modulation of some digestive enzymes and Metabolic processes in developing marine fish: Applications to diet formulation ArchiMer
Zambonino, Jose-luis; Cahu, Chantal.
During these last 20 years, many studies have focussed on the development of the digestive tract in marine fish larvae. Most of the studies aimed at acquiring knowledge on the optimal form of dietary supply for different nutrients, in order to formulate a compound diet able to totally replace live preys in the fish larvae feeding sequence. Consequently, most of the studies aimed at describing the effect of dietary adaptation on digestive enzymes profile, the morphology of the main organs, while others aspects of the physiology of the larvae digestive tract (gut hormones, intestinal transport...) were poorly investigated. This review reports the more recent data on dietary modulation of digestive enzymes with a particular emphasis, when possible, on the...

Dietary neutral lipid level and source in marine fish larvae: Effects on digestive physiology and food intake ArchiMer
Morais, Sofia; Conceicao, L; Ronnestad, I; Koven, W; Cahu, Chantal; Zambonino, Jose-luis; Dinis, M.
A growth depressing effect of high dietary neutral lipid levels in marine fish larvae has been reported. This may be a result of a decrease in the efficiency or activity of digestive enzymes, a reduction in absorption efficiency and/or a decrease in food intake. The present work reviews recent studies carried out on commercially valuable species (Atlantic herring, Senegalese sole, European seabass and gilthead seabream) that investigated the effects of neutral lipid level and lipid source (fatty acid composition) on some of these key factors influencing larval growth. The results seem to collectively indicate that lipid transport from the enterocytes into the body may be more problematic in larval stages dealing with high neutral lipid diets than lipolytic...

Dietary phospholipids are more efficient than neutral lipids for long-chain polyunsaturated fatty acid supply in European sea bass Dicentrarchus labrax larval development ArchiMer
Gisbert, Enric; Villeneuve, Laure; Zambonino, Jose-luis; Quazuguel, Patrick; Cahu, Chantal.
We evaluated the effects of dietary lipid class (phospholipid vs. neutral lipid) and level of n-3 long-chain PUFA (LC-PUFA) on the growth, digestive enzymatic activity, and histological organization of the intestine and liver in European sea bass larvae. Fish were fed from the onset of exogenous feeding at 7 to 37 d post-hatch with five isoproteic and isolipidic compound diets with different levels of EPA and DHA. Diet names indicated the percentage of EPA and DHA contained in the phospholipids (PL) and neutral lipids (NL), as follows: PL5, PL3, PL1, NL1, and NL3. Histological observations showed different patterns of lipid absorption and accumulation in the intestinal mucosa depending on the level and nature of the dietary lipid fraction. Fish fed high...

Dietary protein hydrolysate and trypsin inhibitor effects on digestive capacities and performances during early-stages of spotted wolffish: Suggested mechanisms ArchiMer
Savoie, A.; Le Francois, N. R.; Lamarre, S. G.; Blier, P. U.; Beaulieu, L.; Cahu, Chantal.
Growth rate is dependent upon adequate provision of amino acids especially in newly-hatched fish which experience very high growth rate. The replacement of a fraction of protein content by partially hydrolyzed (pre-digested) proteins was carried out and the digestive capacities and performances of larval/juvenile spotted wolffish (Anarhichas minor) were measured. The goal of this study was to verify whether the scope for growth is principally dictated by the proteolytic capacity of the digestive system by examining the effect of protein hydrolysates (PH) and trypsin inhibitor dietary inclusion on protein digestion/assimilation capacities, growth and survival. Four experimental diets were examined: C (control) I (supplemented with 750mg/kg soybean trypsin...

Dietary TAG source and level affect performance and lipase expression in larval sea bass (Dicentrarchus labrax) ArchiMer
Morais, Sophia; Cahu, Chantal; Zambonino-infante, Jose-luis; Robin, Jean; Ronnestad, I; Dinis, M; Conceicao, L.
The influence of dietary TAG source (fish oil, triolein, and coconut oil) and level (7.5 and 15% of the diet) on growth, lipase activity, and mRNA level was studied in sea bass larvae, from mouth opening until day 24 and from day 37 to 52. Fish oil and triolein induced better growth in both experiments, this being significant at a higher dietary level. Coconut oil significantly decreased growth at the higher level, possibly as the result of an excessive supply of medium-chain TAG. Growth was not related to lipase specific activity, suggesting a production in excess to dietary needs. Body lipid content was positively related to dietary lipid level and was affected by lipid quality. In addition, larval FA composition generally reflected that of the diet. The...
